ClickOnce deployment

My current VB6 application uses something similar to ClickOnce technology
for automatic upgrades. User starts on his (her) workstation not the
application itself, but the utility which compares versions of the main
application EXE on the mapped drive (located on the company server) and EXE
located on the user workstation. If they are different then the utility
replaces an EXE with the server copy and starts it.
I'm starting on the new VB 2005 project and was wondering if ClickOnce
deployment will work for me.
The new application is going to be used by many companies with different
letters for mapped drive - in one company it's N:, in another it's R: and so
on.
Will I need to run Publish Wizard as many times as many different locations
I have on client's servers? Or once I ran the wizard I will get a package
which will be used by all companies. Any adjustment needed?

Does ClickOnce technology supports installations (upgrades) of third party
stuff (all the time when I had to upgrade Crystal Reports, for instance from
v 7 to v 8, or from v10 to v11.5 I had to recreate setup package in
InstallShield and reinstall my app on all workstations manually)? How is it
going to work with ClickOnce technology? The same way-manual reinstallation,
or some automation is used?
