Yes, you should save the last used directory somewhere. Try using
ConfigParser.

last_used_directory = ... # retrieve from saved options; default = "."
filename = tkFileDialog.askopenfilename(initialdir=last_used_ directory)
if filename is not None:
# save the directory: os.path.dirname(filename)
...
