Custom Collections

Hello all,
There are a few ways to implement collections of custom business entities. I have found some good examples where you can create a Base Business Entities collection by creating a common class derving from Collection Base. This seems to work great but I am not sure how best to populate this collection.

Assuming either DataSet, DataTable or DataReader, should I pass these to the collection constructor or should I iterate over the rows and create a business entity and call the add on the collectionbase?

Another question, assuming a relational datastore, should the add method on the collection call the DAL and update the database or should a business workflow do that?

I realize these are general questions, just have not found any good articles on this subject.
