Socket backlog

I have a SMTP server with an async socket listener developed using Vb.Net. Multiple such servers are behind a Load Balanced, bound to a VIP. I sometimes find that a machine will stop accepting connections all together. All operating parameters shcuh as memory, handles, threads etc are normal. Any suggessions on how I could debug the server, or what the issue could be ? My socket backlog parameter is set to 100. What is the significance of that ? Is there a way I can query the queue

Thanks
