mysql table cell before and after data entered

Is there some difference between a mysql table cell before any data has been entered into it (ie. when the row was inserted, this field had blanks entered '' ) and the cell once data has been entered and then subsequently removed from that cell?

The reason I ask is that in the first case, my code acts like it should, but in the latter, it screws it up. Both times there is "nothing" in the cell, but I'm wondering if I've changed the data type or something by doing that.
