NULL

I have table with columns which has NOT NULL definition.
What can I do to be ensure that update or insert will not modify
table with empty values?
MY EXAMPLE:

if($zm=='')
$zm=null ;
or what can I do?

How can I use NOT NULL table column definition?
or what NOT NULL definition is for?
