Machine.config error - help, anyone?

The first few times I am hitting an ASPX page I get the following error
(after that the page works fine w/ no changes - how do I avoid this in the
first place?):
Configuration Error

Description: An error occurred during the processing of a configuration file
required to service this request. Please review the specific error details
below and modify your configuration file appropriately.

Parser Error Message: Access is denied: 'aname'.
and the line is:
Line 198: <add assembly="*"/>
