An ISO file is normally (not always though!) a CD- or DVD-Image. (In the case of SP3, probably CD.) So, you'd have to burn it to a CD. (That doesn't mean "burn a CD with that file on it" but burn a CD from that image. Check

Of course, this could be problematic, if you can't open any programs on your computer any more. If you can't get it burnt on your computer, try doing so from some other system. (By the way, normally you'd just start Windows Update to get SP3...)
However, you should try getting Windows working again, before you attempt to update it. My suggestion would be to boot with the Windows CD and choose the repair option. (Not quite sure of the exact steps now, as it's been a while, but there's a repair option in there somewhere.)
If that doesn't work, a complete reinstall might be something you should consider. Your system seems to be in a pretty broken state.
